The SIRT1 activation claim is contested — Pfizer researchers showed the original assays had a fluorescent artifact. Human bioavailability is poor (<1% without modifications). Sinclair continues to promote it paired with NMN. Micronized formulations improve absorption ~3.6×. No human longevity RCTs. Modest anti-inflammatory and cardiovascular signal in short-term trials.
Early-stage — most claims are extrapolated from animal or in-vitro studies with no human replication at scale. Longevity effects specifically resist short-term human measurement, which means both positive and negative signals are genuinely difficult to read.
