The SIRT1 activation claim is contested — Pfizer researchers showed the original assays had a fluorescent artifact. Human bioavailability is poor (<1% without modifications). Sinclair continues to promote it paired with NMN. Micronized formulations improve absorption ~3.6×. No human longevity RCTs. Modest anti-inflammatory and cardiovascular signal in short-term trials.
The sirtuin activation mechanism is contested at human doses. Poor oral bioavailability without micronized or liposomal form. May act as phytoestrogen at high doses. Mild CYP3A4 inhibitor.
